Type II topoisomerases are ubiquitous and ancient enzymes that control DNA supercoiling by making double-stranded DNA breaks and transporting one segment of DNA through another. Using cryoEM we have determined the basis of the chiral loop formation and T-segment trapping by E. coli gyrase which is a first step in gyrase catalytic cycle.
